外源精胺对黑果枸杞不同性质愈伤组织诱导再生器官及玻璃化的影响

Effects of Exogenous Spermine on the Induction of Organ Regeneration and Vitrification of Callus With Different Natures of Lycium ruthenicum Murr.

  • 摘要: 以黑果枸杞不同性质的愈伤组织为试材,将不同浓度的精胺(Spm)添加于MS培养基中,研究外源Spm对黑果枸杞不同性质愈伤组织诱导再生器官及玻璃化的影响,以期为黑果枸杞植株再生技术、遗传转化等提供参考依据。结果表明:添加适宜浓度的Spm均能显著提高愈伤组织再生率、降低玻璃化率。4种不同性质愈伤组织的再生率、玻璃化率影响效果依次为芽愈伤组织(BEC)>叶愈伤组织(LEC)>茎愈伤组织(SEC)>花药愈伤组织(AEC)。青海诺木洪(QH-N)和宁夏中宁(NX-Z)的BEC添加Spm最佳浓度均为30 mg·L-1,再生率分别达到60.62%和63.89%,均无玻璃化苗发生;LEC添加Spm最佳浓度均为20 mg·L-1,再生率分别达到49.48%和52.48%,均无玻璃化苗发生;SEC添加Spm最佳浓度分别为10 mg·L-1和15 mg·L-1,再生率分别为33.60%和42.11%,玻璃化率分别降至1.14%和0%;AEC最佳浓度均为15 mg·L-1,再生率分别为19.89%和34.86%,玻璃化率分别降至12.19%和8.97%。外源精胺能有效促进愈伤组织再生率,并不同程度减轻或抑制玻璃化苗的发生。

     

    Abstract: In this study,the effects of exogenous Spm on the iduction of regenerative organs and vitrification of callus with different properties of Lycium ruthenicum Murr.by add different concentrations of Spm into MS medium,in order to provide reference for plant regeneration technology and genetic transformation of Lycium ruthenicum Murr.The results showed that the adding of Spm in appropriate concentrations could increase the rate of callus regeneration and reduce the occurance of vitrification significantly.Compared the effects of Spm on four different types of callus,BEC had the best result,followed by LEC,SEC and AEC.For BEC of QH-N and NX-Z,the optimal concentration of Spm was 30 mg·L-1,the regeneration rate was 60.62% and 63.89%,respectively,and there was no vitrified seedling.The optimal concentration of LEC adding Spm was 20 mg·L-1,the regeneration rate reached 49.48%and 52.48%,respectively,and there was no vitrified seedling.When adding Spm in concentration of 10 mg·L-1 and 15 mg·L-1,SCE had regeneration rate of 33.60%and 42.11%,and the vitrification rate was reduced to 1.14%and 0,respectively.For AEC,the best concentration of adding Spm was 15 mg·L-1.The regeneration rate could reach to 19.89% and 34.86%,and the vitrification rate could reduced to 12.19% and 8.97%,respectively.These results indicated that exogenous spermine could promote the regeneration rate of callus,and reduce the occurrence of vitrified seedlings in different degrees.
