黄精属植物SRAP分子标记遗传多样性分析

Genetic Diversity Analysis of Polygonatum Mill.Based on SRAP Molecular Markers

  • 摘要: 以34份黄精属种质资源(多花黄精、湘黄精、早花黄精、湖北黄精、滇黄精、玉竹)为试材,采用SRAP分子标记技术,研究其遗传多样性,以期为黄精属植物种质鉴别和分子育种提供参考依据。结果表明:筛选了12对SRAP多态性引物,共检测到106个位点,多态性百分率为100.0%,Nei′s基因多样性指数(H)平均值为0.285 8,Shannon多样性指数(I)平均值为0.439 9,遗传分化系数Gst为0.536 0,种质间基因流Nm为0.432 8。34份种质间遗传相似系数为0.500 0~0.952 8,平均值为0.669 8。湘黄精和滇黄精种质遗传差异最小,而多花黄精和滇黄精种质遗传差异最大。聚类分析可将34份黄精属种质划分为六大类,多花黄精、湘黄精、早花黄精、湖北黄精、滇黄精、玉竹分别聚为一类。该研究的12对多态性SRAP分子标记能有效鉴别黄精属不同种质,将为黄精属遗传多样性分析、种质鉴定、分子标记辅助育种等提供参考依据。

     

    Abstract: The genetic diversity of 34 germplasm resources of Polygonatum Mill.(Polygonatum cyrtonema,Polygonatum hunanense,Polygonatum praecox,Polygonatum zanlanscianense,Polygonatum kingianum and Polygonatum odoratum) were analyzed by SRAP molecular marker,in order to provide reference for germplamsm identification and molecular breeding of Polygonatum Mill..The results showed that a total of 106 loci were detected using 12 pairs of SRAP polymorphic primers,with a polymorphism percentage of 100.0%.The average Nei’s gene diversity index(H),the average Shannon diversity index(Ⅰ),the genetic differentiation coefficient(Gst),and the gene flow between germplasm was 0.285 8,0.439 9,0.536 0,and 0.432 8,respectively.The genetic similarity coefficient between 34 germplasm ranges from 0.500 0 to 0.952 8,with an average value of 0.669 8.The genetic differences between P.hunanense and P.kingianum germplasm were the smallest,while the genetic differences between P.cyrtonema and P.kingianum germplasm were the largest.Cluster analysis divided 34 germplasm of Polygonatum Mill.into 6 major categories,with P.cyrtonema,P.hunanense,P.praecox,P.zanlanscianense,P.kingianum, and P.odoratum clustered into one category,respectively.The 12 pairs of polymorphic SRAP molecular markers in this study could effectively identify different germplasm of Polygonatum Mill,and provide reference for genetic diversity analysis,germplasm identification,and molecular marker assisted breeding of Polygonatum species.
