番茄抗颈腐根腐病基因Frl的定位分析

Mapping Analysis of Tomato Fusarium Crown and Root Rot Resistance Gene Frl

  • 摘要: 以含有番茄颈腐根腐病抗病基因Frl的番茄材料19912与感病材料Moneymaker构建F2群体,采用父母本的重测序技术结合BSA的关联分析方法,进一步对番茄颈腐根腐病抗病基因Frl候选区间关联分析,以期筛选得到抗性基因的候选区间。结果表明:通过对父母本的重测序及BSA混池关联分析,将Frl基因定位于1号染色体的2.63~81.19 Mb区间;利用SNP分型技术,结合BSA的测序结果,将区间缩小至17.4~23.5 Mb。该研究不仅可以为抗病基因的克隆及应用提供参考依据,还将为解析番茄颈腐根腐病抗性机制奠定基础。

     

    Abstract: F2 population was constructed using tomato material 19912 containing the resistance gene Frl to tomato neck rot root rot and susceptible material Moneymaker.The association analysis method of BSA was used to further analyze the candidate intervals of the resistance gene Frl in tomato fusarium crown and root rot, with the aim of screening candidate intervals for resistance genes.The results showed that by resequencing the father and mother parents and analyzing the mixed pool association of BSA,the Frl gene was located in the 2.63-81.19 Mb interval of chromosome 1.By using SNP typing technology and combining with the sequencing results of BSA,the interval was reduced to 17.4-23.5 Mb.This study will not only provide a basis for the cloning and application of disease resistance genes, but also lay a foundation for analyzing the resistance mechanism of tomato neck rot root rot disease.
