MsRCI2A、MsRCI2B、MsRCI2C基因超量表达对紫花苜蓿耐冷的影响
Effects of Overexpression of MsRCI2A,MsRCI2B and MsRCI2C Genes on Cold Tolerance of Alfalfa

摘要: 以紫花苜蓿为试材,对紫花苜蓿来源的RCI2类基因MsRCI2A/2B/2C在4℃的表达差异进行分析,采用农杆菌介导的方法对紫花苜蓿进行遗传转化,研究了超量表达MsRCI2A/2B/2C基因对紫花苜耐冷性的影响,以期为培育耐冷紫花苜蓿提供参考依据。结果表明:MsRCI2A/2B/2C基因均受低温诱导,转基因苜蓿材料经4℃冷处理12 d后,叶绿素相对含量、相对电导率、丙二醛含量才发生明显变化,对冷的敏感性下降。并且4℃处理后,转基因株系较WT(野生型)积累更多脯氨酸和可溶性糖,抗氧化酶(SOD、POD和CAT)活性在低温6 d时迅速上升,响应冷胁迫更迅速。综上所述,MsRCI2A/2B/2C基因的超量表达提高了紫花苜蓿的耐冷性。Abstract: Alfalfa was used as the test material.The expression differences of RCI2 genes MsRCI2 A/2 B/2 Cderived from alfalfa were analyzed at 4 ℃,and then the genetic transformation of alfalfa was carried out by agrobacterium-mediated method.The effect of overexpression of MsRCI2 A/2 B/2 C gene on cold tolerance of alfalfa,in order to provide reference for cultivating cold tolerant alfalfa.The results showed that the MsRCI2 A/2 B/2 C genes were all induced by 4 ℃.The chlorophyll relative content,relative conductivity,and MDA content of the transgenic alfalfa changed significantly after12 days of 4℃treatment,indicating that their sensitivity to 4℃ was reduced.And after treatment at4℃,the transgenic plants accumulated more proline and soluble sugar than WT plants,and the activities of antioxidant enzymes(SOD,POD and CAT)increased rapidly at 6 days of treatment at4℃,and responded to cold stress more rapidly.In summary,the overexpression of MsRCI2 A/2 B/2 C genes improved the cold tolerance of alfalfa.
