无刺刺梨茎段快繁体系的建立

Establishment of Rapid Propagation System for Stem Segments of Rosa roxburghii f.eseiosa ‘Wuci 2’

  • 摘要: 以“无刺2号”当年生营养枝为外植体,采用不同浓度的植物生长调节剂进行组培试验,研究了不同消毒方式、激素浓度对外植体消毒效果、初代培养、增殖培养及生根培养的影响,以期建立较为成熟的无刺刺梨组织培养体系。结果表明:外植体经清水冲洗后用75%的酒精消毒15 s,以0.1%的升汞消毒8 min,存活率为85.28%;受污染后的植株在超声洗涤液中洗涤,洗涤液中添加多菌灵60 mg·L-1和青霉素40 mg·L-1,同时配置培养基时添加多菌灵60 mg·L-1和青霉素40 mg·L-1是最佳处理;适合“无刺2号”初代培养的培养基为MS+1 mg·L-1 6-BA+0.1 mg·L-1 NAA,萌芽率达92.04%;增殖培养基为MS+0.2 mg·L-1 6-BA+0.1 mg·L-1 NAA,增殖系数为3.9;在1/2MS+0.1 mg·L-1 IBA+20 g·L-1蔗糖+0.5 g·L-1活性炭的培养基上,“无刺2号”的生根率为93.17%。研究结果对“无刺2号”的苗木产业化生产和种质资源保存具有重要意义。

     

    Abstract: The current year vegetative shoots of ‘Wuci 2’ were used as explants, the effects of different disinfection methods and hormone concentration on the disinfection effect of explants, primary culture, proliferation culture and rooting culture were studied, in order to establish tissue culture system of Rosa roxburghii f.eseiosa Ku.The results showed that the survival rate was 85.28% after the explants were washed with water and disinfected with 75% alcohol for 15 seconds and 0.1% HgCl2 for 8 minutes.The contaminated plants were washed in ultrasonic washing solution, adding carbendazim 60 mg·L-1 and penicillin 40 mg·L-1 in the washing solution, and adding carbendazim 60 mg·L-1 and penicillin 40 mg·L-1 in the medium was the best treatment.The suitable medium for the primary culture of ‘Wuci 2’ was MS+1 mg·L-1 6-BA+0.1 mg·L-1 NAA,and the germination rate reached 92.04%.The suitable proliferation culture medium was MS+0.2 mg·L-1 6-BA+0.1 mg·L-1 NAA,and the proliferation coefficient was 3.9.The rooting rate of ‘Wuci 2’ was 93.17% on the medium of 1/2MS+0.1 mg·L-1 IBA+20 g·L-1 sucrose+0.5 g·L-1 activated carbon.The results of this study were of great significance for the industrial production and conservation of germplasm resources of ‘Wuci 2’.
