大花蕙兰‘大凤’×豆瓣兰‘太极圣梅’ F1代试管开花诱导

Flowering Induction of F1 Generation of Cymbidium hybridium ‘Dafeng’×C.serratum var.goeringii ‘Taijishengmei’ in vitro

  • 摘要: 以大花蕙兰‘大凤’×豆瓣兰‘太极圣梅’(Cymbidium hybridium‘Dafeng’×C.serratum var.goeringii‘Taijishengmei’)F1代试管苗为试材,采用PP333或CCC预处理,添加不同浓度的6-BA,测定植株形态和生理指标,建立试管开花诱导体系,缩短兰花植株生长的童期,以期为兰花高效育种提供参考依据。结果表明:不同浓度PP333或CCC预处理抑制了植株株高、根数和鲜质量的增加,但促进叶片分裂;试管苗预处理培养于1/2MS+PP333 1.0 mg·L-1+香蕉泥120 g·L-1+蔗糖30 g·L-1+琼脂8 g·L-1+活性炭0.5 g·L-1的培养基中30 d,再转接到1/2MS+NAA 0.2 mg·L-1+香蕉泥120 g·L-1+蔗糖30 g·L-1+琼脂8 g·L-1+活性炭0.5 g·L-1的培养基可诱导试管开花,花芽诱导率为6.67%,但花芽后期未顺利开放;PP333或CCC预处理下试管苗植株可溶性糖和可溶性蛋白质含量均高于CK,并促进C/N值的提高以及植株全氮含量的降低。

     

    Abstract: The F1 generation plantlets of Cymbidium hybridium ‘Dafeng’ × C.serratum var.goeringii ‘Taijishengmei’ were used as test material pretreated with PP333 or CCC,and subsequently added with different concentrations of 6-BA to determine plant morphology and physiological indicators, a test-tube flowering induction system was established and the childhood period of orchid plants was shortened, in order to provide reference for breeding of orchid.The results demonstrated that pretreatment with different concentrations of PP333 or CCC inhibited the increasing of plant height, root number and fresh weight, but promoted leaf division.The test tube seedlings were pre-cultured in 1/2MS+PP333 1.0 mg·L-1+banana puree 120 g·L-1+sucrose 30 g·L-1+agar 8 g·L-1+activated carbon 0.5 g·L-1 for 30 days.Then, the culture medium of 1/2MS+NAA 0.2 mg·L-1+banana pulp 120 g·L-1+sucrose 30 g·L-1+agar 8 g·L-1+activated carbon 0.5 g·L-1 could induce the flower bud flowering, the flower bud induction rate was 6.67%,but the flower bud did not open successfully in the late stage.The contents of soluble sugar and soluble protein were higher than CK under PP333 or CCC pretreatment, which promoted the increase of C/N value and the decrease of total nitrogen content.
