红托竹荪液体菌种培养基及培养条件优化

Optimization for the Medium for Liquid Spawn and Culture Conditions of Dictyophora rubrovolvata

  • 摘要: 以红托竹荪菌种为试材,采用液体摇瓶培养方法,研究了红托竹荪液体培养基的碳氮源筛选和培养条件优化,以期为红托竹荪液体菌种的制备和扩大培养提供参考依据。结果表明:最佳碳源为葡萄糖,最佳氮源为蛋白胨,最佳配方为葡萄糖30 g·L-1,蛋白胨1.5 g·L-1,KH2PO4 2 g·L-1,MgSO4 1 g·L-1,(NH42SO4 1 g·L-1,维生素C 0.016 g·L-1,生长素(IAA)0.001 6 g·L-1。最佳培养条件为25℃,摇床转速150 r·min-1,培养天数20 d。扩大培养的最佳培养条件为装液量每1 000 mL装500 mL,接种量15%,培养天数15 d,从一级种制备到扩大培养共计35 d。

     

    Abstract: Taking Dictyophora rubrovolvata as test material, the carbon and nitrogen source selection and culture condition optimization of its liquid medium of Dictyophora rubrovolvata were studied through liquid shake flask culture method, in order to provide a reference for the preparation and extended culture of liquid strains of Dictyophora rubrovolvata.The results showed that glucose and peptone were the optimal carbon source and nitrogen source, respectively.The optimal formula was 30 g·L-1 of glucose, 1.5 g·L-1of peptone, 2 g·L-1of KH2PO4,1 g·L-1of MgSO4,1 g·L-1 of(NH42SO4,0.016 g·L-1 of vitamin C,and 0.0016 g·L-1 of indole acetic acid(IAA).Besides, the optimal culture conditions were 25 ℃,150 r·min-1 of shaking speed, and 20 days of culture period.The optimal culture conditions were as follows 500 mL of liquid per 1 000 mL of culture volume, 15% of inoculation size, and 15 days of culture period; 35 days were required from first-degree species preparation to the extended culture.
