不同基质对桑黄菌丝生长及酶活性的影响

Effects of Different Substrates on Mycelial Growth and Enzyme Activity of Pellinus linteus

  • 摘要: 以桑黄为试材,利用粗木屑、细木屑、小麦秸秆、花生秸秆、玉米秸秆、油菜秸秆和大豆秸秆7种主料对桑黄进行培养,采用单纯型格子法进行混料设计,筛选以粗木屑、油菜秸秆和大豆秸秆为主料的配方,对菌丝生长速率、漆酶、多酚氧化酶、纤维素酶、半纤维素酶以及酸性蛋白酶活性进行了测定,分析胞外酶活性与菌丝生长速率之间的相关性,优化适合桑黄生长的配方,以期为桑黄培养配方的筛选提供参考依据。结果表明:不同基质对各胞外酶活性大小有不同的影响,且酶活性大小与菌丝生长速率之间的偶联性较差。优化得出适合桑黄生长的配方为粗木屑41.5%、油菜秸秆41.9%、大豆秸秆16.6%、麦麸20%、蔗糖1%、碳酸钙1%。

     

    Abstract: Pellinus linteus was used as the test material.Seven main materials, namely coarse sawdust, sawdust, wheat straw, peanut straw, corn straw, rape straw and soybean straw were used for Pellinus linteus cultivation.The simple lattice method for mixed material design was used.The formula that main materials inclued coarse sawdust, rape straw and soybean straw was screened.The mycelial growth rate and enzyme activities of laccase, polyphenol oxidase, cellulase, hemicellulase and acid protease were measured.The correlation between mycelial growth rate and extracellular enzymes activities were analyzed, in order to provide a reference for screening the formula of Pellinus linteus cultivation.The results showed that different substrates had different effects for the extracellular enzymes activities.And the contingency between extracellular enzymes activities and mycelial growth rate was poorer.The optimized formula included 41.5% coarse sawdust, 41.9% rape straw, 16.6% soybean straw, 20% wheat bran, 1% sucrose, 1% calcium carbonate.
