利用ARTP诱变技术选育白灵菇高产菌株

Breeding High-yield Strain of Pleurotus nebrodensis by ARTP Mutagenesis Technology

  • 摘要: 以白灵菇“中农1号”为试材,研究了白灵菇“中农1号”原生质体制备条件;以不做处理的“中农1号”为CK,首次采用常压室温等离子体(atmospheric room temperature plasma, ARTP)诱变技术,研究了其对菌株诱变后子实体产量的影响,以期获取相比CK菌株产量更高的优良菌株。结果表明:通过单因素试验得到白灵菇“中农1号”原生质体的最佳制备条件为酶解温度28℃、酶解时间3 h、溶壁酶浓度为20 mg·mL-1、渗稳剂使用0.6 mmol·L-1 KCl。ARTP诱变最佳时间为60 s,致死率为94%时正突变率最为稳定。通过平板筛选试验初筛结果显示,生长速度快、产漆酶、纤维素酶能力强的菌株有4株,进一步通过了LBL脱色试验复筛,筛选出结实性好的4株菌株ZN-37、ZN-20、ZN-39、ZN-19。通过测定白灵菇栽培产量确定最终的高产菌株为ZN-19,总产量高达966.95g,相比CK菌株总产量多37%。

     

    Abstract: The protoplast preparation conditions of Pleurotus nebrodensis ’Zhongnong No.1’ were studied by using Pleurotus nebrodensis ’Zhongnong No.1’ as test material.The a Zhongnong No.1’without treatment was used as CK,and the atmospheric room temperature plasma(ARTP)mutagenesis technique was used for the first time to study its effect on the yield of fruiting bodies after mutagenesis,in order to obtain superior strains with higher yield than CK.The results showed that the optimum preparation conditions of protoplasts of Pleurotus nebrodensis ’Zhongnong No.1’obtained by single factor test were as follows,enzymolysis temperature 28 ℃,enzymolysis time3 hours,lywallzyme concentration 20 mg·mL-1,osmotic stabilizer 0.6 mmol·L-1 KCl.The optimal time of ARTP mutagenesis was 60 seconds,and the positive mutation rate was the most stable when the lethal rate was 94%.The preliminary screening results of the plate screening test showed that there were 4 strains with fast growth rate,strong laccase and cellulase production ability.Further,the LBL decolorization test was rescreened,and 4 strains ZN-37,ZN-20,ZN-39 and ZN-19with good seed setting were screened out.By measuring the cultivation yield of Pleurotus nebrodensis,the final high-yield strain was determined to be ZN-19,with a total yield of 966.95 g,which was 37 %more than the total yield of the CK group.
