基于花脸香蘑基因组的SSR分子标记开发

Development of SSR Molecular Markers Based on Whole Genome of Lepista sordida

  • 摘要: 以4个花脸香蘑菌株及已公布的花脸香蘑基因组为试材,采用生物信息学和试验验证相结合的方法,研究了花脸香蘑基因组上的SSR类型及分布,设计了50对SSR引物开展试验验证,以期为花脸香蘑的种质资源鉴定、品种选育、分子标记辅助育种等研究提供参考依据。结果表明:总计检索到7种SSR类型的2 486条序列,其中主要以单碱基(P1)类型为主,占总SSR的67.70%,且分布密度较高;四、五、六碱基重复的类型较多,但数量较少。由验证试验获得4对多态性高、重复性好、特异性好的引物,分别为HLMSSR001、HLMSSR004、HLMSSR015和HLMSSR041。其中,HLMSSR001和HLMSSR004可以通过单一的引物扩增即可将4个花脸香蘑菌株进行区分鉴定。同时,基于4对引物进行花脸香蘑DNA指纹编码的构建,获得了4个花脸香蘑菌株唯一的8位数分子指纹数据库编码。

     

    Abstract: Based on experimental validation and bioinformatics methods, SSR analysis result were exploited from the genomic of Lepista sordida and 4 strains.In the meantime, 50 SSR were selected for primer design and experimental verification,in order to provide theoretical basis for the germplasm resources identification, variety selection and molecular marker-assisted breeding of L.sordida.The results indicated that L.sordida had 7 SSR types.A total of 2 486 SSR were retrieved, P1 type was the main type, accounting for 67.70% and also higher distribution density than other type.There were more types of four, five and six base repeats.However, the number was small.Four primers with high pair property, repeatability and specificity were obtained from the validation experiment, which were HLMSSR001,HLMSSR004,HLMSSR015 and HLMSSR041.HLMSSR001 and HLMSSR004 could be amplified by a single primer to distinguish and identify the 4 strains of L.sordida.At the same time, the DNA fingerprint coding of 4 pairs of primers was constructed and the unique 8-digit molecular fingerprint database codes of 4 L.sordida strains were obtained.
