Abstract:
The effects of acidity of the extract,the amount of activated clay,the treatment temperature and time on the clearance efficiency of turbidity and the recovery of polysaccharide were investigated by using the extract of polysaccharides fromAuricularia auriculariaand evaluation with turbidimetry and phenol-sulfuric acid methods.Box-Behnken experiment design was employed to establish and analyze the mathematical model of the influence of each factor on the removal efficiency of turbidity and the recovery of crude polysaccharide,in order to provide reference for the rational development and utilization of the extract of crude polysaccharides from Auricularia auricularia.The results showed that the protein content in the extract was reduced from 0.39 mg·mL-1 to 0.05 mg·mL
-1 after treatment with activated clay.The optimal process conditions for removal of the turbidity in the extract were as follows,the recovery of polysaccharides and removal efficiency of turbidity were85.7%and 61.0%respectively by addition of activated clay at a solid-liquid ratio of 1.86g·mL
-1 to the extract(pH 7.0)and treatment at 44.8℃for 61.2 minutes.
