Abstract:
The embryoids obtained from garden asparagus pollen grain culture were used as experimental materials.In the adventitious buds, MS+sucrose 30 g·L
-1+agar powder 8 g·L
-1 were used as the basic medium, pyrimidinol(0,0.05,0.10,0.50,1.00,2.00 mg·L
-1) and naphthylacetic acid(NAA)(0,0.05,0.10,0.20,0.50,1.00 mg·L
-1) with different mass concentrations were added, respectively.The effects of pyrimidinol and NAA concentrations on the growth of regenerated plants with different genotypes were studied, in order to optimize and improve the propagation efficiency of garden asparagus(Asparagus officinalis L.) plants in vitro.The results showed that the net growth height of regenerated plants treated with 0.50 mg·L
-1 pyrimidinol in the medium increased by 1.05 cm and 1 root respectively, and the root system became longer and the lateral roots increased.After adding 0.50 mg·L
-1 pyrimidinol and 0.20 mg·L
-1 NAA to MS medium, the net growth height of regenerated plants, the number of stems increased by 1.60 cm and 2.0 roots, respectively, and the root system grew vigorously.And there was no significant difference among the genotypes in the promoting effect of 0.50 mg·L
-1 pyrimidinol and 0.20 mg·L
-1 NAA on plant regeneration from garden asparagus embryoids.It was speculated that adding 0.50 mg·L
-1 pyrimidinol and 0.20 mg·L
-1 NAA to MS medium could improve the reproductive efficiency of regenerated plants from garden asparagus embryoids.
