不同基因型甜瓜再生体系的比较

Comparison of Regeneration System of Melon With Different Genotypes

  • 摘要: 以厚皮甜瓜品种“雪彤8号”和“雪彤9号”、薄皮甜瓜品种“翠玉6号”和“珍甜18”为试验材料,采用甜瓜离体子叶再生的方法,研究了不同激素及浓度配比对不同基因型甜瓜子叶再生体系的影响,以期为甜瓜的遗传转化和基因工程育种提供参考依据。结果表明:甜瓜种子最适宜的NaClO消毒浓度为0.2%,利用MS培养基添加细胞分裂素6-BA和生长素IAA或NAA诱导甜瓜子叶产生簇生芽,厚皮甜瓜和薄皮甜瓜在诱导芽分化上存在明显差异,厚皮甜瓜“雪彤8号”和“雪彤9号”最适宜的芽分化培养基为MS+1 mg·L-1 6-BA+0.1 mg·L-1 IAA,芽分化率分别可达90.3%和88.9%,而薄皮甜瓜“翠玉6号”和“珍甜18”最适宜的芽分化培养基为MS+2 mg·L-1 6-BA+0.3 mg·L-1 IAA,芽分化率分别可达85.5%和91.7%。厚皮甜瓜和薄皮甜瓜最佳的芽伸长培养基一致为MS+0.2 mg·L-1 KT,芽伸长快且长势好。最佳的生根培养基为1/2MS+0.1 mg·L-1 IAA,生根快且粗壮。

     

    Abstract: Thick-skin muskmelon varieties ‘Xuetong 8’ and ‘Xuetong 9’ and thin-skin muskmelon varieties ‘Cuiyu 6’ and ‘Zhentian 18’ were used as experimental materials with the method of regeneration of melon cotyledon in vitro.The effects of different hormones and concentration ratio on the regeneration system of melon cotyledon were studied, in order to provide reference for genetic transformation and genetic engineering breeding of melon.The results showed that the optimal disinfection concentration of NaClO solution was 0.2% for melon seeds, and the cell kinin 6-BA and auxin IAA or NAA were added into MS medium to induce the cluster buds of melon cotyledons.There were significant differences in bud differentiation between thick-skin melon and thin-skin melon.The optimal medium for bud differentiation of ‘Xuetong 8’ and ‘Xuetong 9’ was MS+1 mg·L-1 6-BA+0.1 mg·L-1 IAA,and the bud differentiation rate was 90.3% and 88.9% respectively.While the optimal medium for bud differentiation of ‘Cuiyu 6’ and ‘Zhentian 18’ was MS+2 mg·L-1 6-BA+0.3 mg·L-1 IAA,and the bud differentiation rate was 85.5% and 91.7% respectively.The best medium of bud elongation for thick-skin melon and thin-skin melon was the same which was MS+0.2 mg·L-1 KT,and the shoots elongated quickly and geminated well.The best rooting medium was 1/2MS+0.1 mg·L-1 IAA,and the roots grew quickly and strong.
