黄瓜枯萎病拮抗芽孢杆菌A7-3-14的筛选及鉴定

摘要: 以从敦煌地区盐碱土壤中分离获得的19株细菌为试材,采用平板对峙法和菌丝生长速率法进行筛选,通过菌株形态特征、生理生化试验和16S rDNA序列分析进行鉴定,并采用盆栽试验测定所筛选的拮抗菌,研究了其对黄瓜枯萎病病原菌Fusarium oxysporum的防效,以期得到对黄瓜枯萎病病原菌具有较强拮抗作用的芽孢杆菌。结果表明:从敦煌地区盐碱地土壤中共分离出19株拮抗细菌,其中菌株A7-3-14拮抗作用最强,抑制率为79.22%,菌株鉴定为内生芽孢杆菌Bacillus endophyticus,盆栽防效结果表明该菌株发酵液对黄瓜枯萎病具有显著的防治效果,其处理后的黄瓜苗病情指数为28.14,防治效果达到69.77%。表明分离到的芽孢杆菌菌株A7-3-14可以用于黄瓜枯萎病的生物防治,且具有良好的效果。Abstract: Taking 19 bacteria obtained from the saline-alkali soil in Dunhuang area as test materials, they were selected by the plate confrontation method and myphelium growth rate method, and were identified by the morphological characteristics, physiology and biochemical test and 16 S rDNA sequence analysis, and the antagonistic bacteria were selected by potted plant test, in order to obtain the strong antagonism of cucumber wilt pathogens.The results showed that 19 antagonist bacteria were isolated from saline-alkali soil in Dunhuang, with the inhibition rate was 79.22%.The identification of endogenous Bacillus endophyticus,showed that the fermentation liquid had significant control effect on cucumber blight.The condition index of cucumber seedlings was 28.14 and the control effect reached 69.77%.It was showed that the isolated Bacillus strain A7-3-14 can be used for biological control of cucumber blight and has good results.
