不同温度光照下兰州百合鳞片气培效果

Different Temperature and Light Treatments on the Reproductive Effect of Lilium davidii var. unicolor Scales by Air Culture

  • 摘要: 以兰州百合鳞片试材,采用气培法进行单因素筛选试验及双因素随机区组试验,研究了不同温度、光照对兰州百合鳞片繁殖小鳞茎效果的影响,以期筛选兰州百合鳞片气培最适宜的环境因子。结果表明:不同气培温度条件下催培效果(腐烂率、生根率、分化率)以及小鳞茎催培效果(分化数、生根数、小鳞茎质量、小鳞茎分级)中以25℃处理的催培效果最好,其中鳞片分化率最高达到89.81%,小鳞茎分化数为146.48粒·(100片)-1,20℃处理次之,小鳞茎分化数达到146.67粒·(100片)-1;25、20℃处理的鳞片生根率均达到60.0%以上。双因素随机区组试验结果以A2B1处理下小鳞茎分化数最高,达到278粒·(100片)-1,生根数最多为424.33条·(100片)-1。其中温度对分化率、生根率、小鳞茎质量、小鳞茎分级(>4 mm)的影响均显著。光照对小鳞茎分化数影响显著且对小鳞茎质量影响极显著。通过催培指标间的相关性分析表明,分化率与腐烂率呈显著负相关,生根率与分化率、分化数呈极显著正相关,生根数与分化数、生根率呈极显著正相关;主成分分析中前2个主成分累计方差贡献率达到86.087 6%,其中分化率、分化数、生根率、小鳞茎质量可以作为主要评价指标;对8种温度光照组合下的气培效果进行综合评价得分最高的处理为A2B1。20~25℃为鳞片催培小鳞茎的适宜温度,同时25℃、500 lx环境下鳞片催培效果最佳,可在利用气培法进行兰州百合鳞片规模化繁殖生产中推广应用。

     

    Abstract: Taking the scale of Lilium davidii var. unicolor test materials, the air culture method was selected for single-factor screening test and two-factor randomized block test, to explore the effect of Lilium davidii var. unicolor scales on the reproduction of bulblet under different temperature and light, in order to screen the most suitable Lilium davidii var. unicolor scales for air culture environmental factors.The results showed that the best culture effect(decay rate, rooting rate, differentiation rate) and bulblet culture effect(differentiation number, rooting number, bulblet quality, bulblet grading) under different aeroponic temperature conditions was achieved by 25 ℃ treatment, in which the highest rate of scale differentiation reached 89.81% and the differentiation number of bulblet was 146.48 grains·(100 pieces)-1,followed by 20 ℃ treatment, in which the differentiation number of bulblet reached 146.67 grains·(100 pieces)-1.The rooting rate of scales at 25 ℃ and 20 ℃ reached 60.0%.The results of the two-factor randomized group test showed that the highest number of bulblet differentiation was 278 grains·(100 pieces)-1 and the highest number of roots was 424.33 strips·(100 pieces)-1 under the A2B1 combination, in which the effects of temperature on differentiation rate, rooting rate, bulblet mass and bulblet grading(>4 mm) were significant.The effect of light on the number of bulblet differentiated was significant and the effect on the quality of bulblet was highly significant.Correlation analysis among aeroponics indicators showed that differentiation rate was negatively correlated with decay rate, while rooting rate was positively correlated with differentiation rate and differentiation number, and rooting number was positively correlated with differentiation number and rooting rate.The cumulative variance contribution of the first two components in the principal component analysis reached 86.087 6%,among which differentiation rate, differentiation number, rooting rate and bulblet mass could be used as the main evaluation index.The aeroponic effect of eight temperature and light combinations was evaluated and the highest score was obtained from combination A2B1.20-25 ℃ was the most suitable temperature for bulb cultivation and the best aeroponic effect was obtained at 25 ℃ and 500 lx, which can be promoted and applied in the large-scale propagation and production of Lilium davidii var. unicolor using aeroponics.
