桔梗总黄酮提取工艺优化及其体外抗氧化性

Optimization of Extraction Process and Antioxidant Activity of Total Flavonoids From Platycodonis radix

  • 摘要: 以桔梗总黄酮为试材,采用Plackett-Burman和中心组合设计(central composite design, CCD)优化桔梗总黄酮的提取工艺,使用3种抗氧化体系,研究其体外抗氧化活性,以期为桔梗总黄酮的开发和应用提供参考依据。结果表明:桔梗总黄酮的最佳提取工艺条件为乙醇浓度63%,液料比22∶1 mL·g-1,浸泡时间63 min,超声提取10 min,总黄酮实际得率为0.45%,纯度为58.22%。桔梗总黄酮和维生素C对DPPH·的IC50值分别为0.546 mg·mL-1和8μg·mL-1,对·OH的IC50值分别为0.196 mg·mL-1和0.368 mg·mL-1,总抗氧化能力与维生素C无显著差异。

     

    Abstract: The total flavonoids from Platycodonis radix was selected as research object.Plackett-Burman design and central composite design was used to optimize the extraction process, antioxidant activities were evaluated by three in vitro antioxidant assay, in order to provide a reference for the development and application of flavonoids from Platycodonis radix.The results showed that optimal extraction parameters were ethanol concentration 63%,liquid-solid ratio 22∶1 mL·g-1,soak time 63 minutes and ultrasonic time 10 minutes.Under these conditions, the extraction yield was 0.45%and the purity was 58.22%.The IC50 value of total flavonoids from Platycodonis radix and vitamin C for DPPH·was 0.546 mg·mL-1 and 8 μg·mL-1 respectively, for ·OH was 0.196 mg·mL-1and 0.368 mg·mL-1 respectively, total antioxidant capacity of Platycodonis radix was not significantly different from vitamin C.
