CaASR3基因沉默对辣椒抗旱性的影响

Effects of CaASR3 Gene Silencing on Drought Resistance in Pepper

  • 摘要: 以‘遵辣1号’辣椒为试材,利用VIGS技术获得CaASR3基因沉默后植株,研究了CaASR3基因在干旱胁迫响应中的作用,以期为辣椒抗旱机理的研究提供分子基础。结果表明:基因沉默后,沉默植株CaASR3基因表达量在0、3、6、12 h显著低于对照植株;干旱处理后,CaASR3基因沉默植株叶片比对照植株失水萎蔫严重,过氧化物酶活性、脯氨酸含量、可溶性糖含量以及相对电导率显著上升,相对含水量显著降低,说明沉默CaASR3基因导致辣椒耐旱能力减弱。因此,CaASR3基因参与了辣椒抗旱调节途径。

     

    Abstract: Taking ‘Zunla-1’pepper as test materials,the silencing of CaASR3 pepper plants was approached by using virus-induced gene silencing(VIGS)and the role of this gene in regulating responses to drought was investigated,in order to provide reference for further study of drought resistance mechanism in pepper.The results showed that the expression level of CaASR3 was significantly decreased following CaASR3 silencing at 0,3,6,12hours.After drought treatment,in contrast to control plants,the leaves of CaASR3-silenced plants showed more water loss,peroxidase activity,proline content,soluble sugar content and relative conductivity increased significantly,and the relative water content decreased significantly,indicated that the CaASR3 silencing resulted in the attenuation of drought tolerance in pepper.Thus,the CaASR3 gene was involved in the regulation of drought resistance pathway in pepper.This study lay a foundation for improving drought tolerance of pepper in molecular breeding.
