斑玉蕈木质素降解酶高活性菌株的筛选和鉴定

Screening and Identification of Strain With High Activity of Ligninolytic Enzyme From Hypsizigus marmoreus

  • 摘要: 以前期试验获得的4株斑玉蕈(Hypsizigus marmoreus)杂交菌株为试材,利用亮蓝-PDA平板显色试验、苯胺蓝-PDA平板脱色试验、体细胞不相容试验和ISSR分子标记技术,筛选斑玉蕈木质素降解酶高活性菌株,并鉴定其生物学和遗传学特性,以期丰富斑玉蕈优良品种。结果表明:杂交菌株JZB2124031的木质素降解酶活性最高,亮蓝-PDA平板和苯胺蓝-PDA平板可分别出现明显的变色圈和脱色圈,与其它菌株均可产生拮抗反应。15条ISSR分子标记引物具有扩增稳定性,当相似系数为0.75时,22株菌可以划分为九大类,其中杂交菌株JZB2124031独为一类。综上,杂交菌株JZB2124031具有较高的木质素降解酶活性,与其它供试菌株的亲缘关系存在较大的差异。

     

    Abstract: Four hybrid strains of Hypsizigus marmoreus obtained in previous experiments were used as the main test materials.The superior strain of Hypsizigus marmoreus with high activity of ligninolytic enzyme were screened and their biological and genetic characteristics were identified by remazol brilliant blue R-PDA plate chromogenic test, aniline blue-PDA plate decolorization test, somatic cell incompatibility test and ISSR(inter-simple sequence repeat) molecular marker technology, in order to enrich excellent varieties of Hypsizigus marmoreus.The results showed that the ligninolytic enzyme activity of hybrid strain JZB2124031 was the highest.Remazol brilliant blue R-PDA plate and aniline blue-PDA plate could appear obvious discoloration ring and decolorization ring, respectively.It could produce antagonistic reaction with other strains.The 15 ISSR molecular marker primers showed amplification stability.When the similarity coefficient was 0.75,the 22 strains could be divided into nine groups.Among them, the hybrid strain JZB2124031 was the only one group.In conclusion, the hybrid strain JZB2124031 had a high activity of ligninolytic enzyme and great difference in genetic relationship with other test strains.
