Abstract:
The chloroplast DNA was extracted by CTAB method from the young leaves of Carum buriaticum Turcz.,and the whole chloroplast genome sequence(153 697bp)of C.buriaticum Turcz.was determined and analyzed,in order to study the characteristics of chloroplast microsatellite(cpSSR)of C.buriaticum Turcz.chloroplast genome,and to provide theoretical reference for the molecular identification and genetic diversity of C.buriaticum Turcz.resources.The results showed that a total of 183SSR loci were identified by MISA software in the chloroplast genome sequence of C.buriaticum Turcz.and the average distribution distance between loci was 839.90bp.cpSSR was mainly distributed in the large single-copy region(LSC),and the number of cpSSR in this region was109,accounting for 59.60% of the total.The most common repeat type of the cpSSR was mononucleotide repeat,accounting for 57.69%.The length of cpSSR distributed from 8-15bp and accounted for 78.69%.There were 31cpSSR of length greater than 15bp,indicating that the cpSSR of C.buriaticum Turcz.had the potency of high polymorphism.The cpSSR loci in chloroplast genome of C.buriaticum Turcz.had potential of polymorphism.
