Abstract:
Taking Stevia rebaudiana leaf callus as mutant materials, a salt-tolerant mutant material was created and screened.The callus of Stevia rebaudiana leaves was induced by
60Co-γ ray radiation, and the high-salt medium was used for preliminary screening.Then 100 mmol·L
-1 NaCl, 60 mmol·L
-1 Na
2SO
4 and 70 mmol·L
-1 NaHCO
3 nutrient solution were used to treat the material after the initial screening for 20 days under water culture salt stress.The plant height, stem diameter, internode length, leaf fresh weight, stem fresh weight, leaf dry weight, stem dry weight, leaf length, leaf width and other major agronomic traits were measured, and the content of glycosides of various types was analyzed, in order to provide reference for the breeding of exceltent varieties of Stevia rebaudiana Bertoni.The results showed that after radiation mutagenesis, 13 regenerated seedlings were successfully obtained, 9 salt-tolerant materials were obtained through preliminary salt-tolerant screening, and 7 salt-tolerant mutant materials were obtained after re-screening.No.1-3 were NaCl-tolerant plant, No.4-7 were Na
2SO
4-tolerant plant, and no NaHCO
3-tolerant mutant plant had been obtained.The agronomic characters and glucoside contents of 7 Rinds salt-tolerant mutant materials all showed varying degrees of variation, and finally a NaCl-tolerant mutant material with excellent comprehensive properties was obtained.Through the creation and screening of mutants, 7 new salt tolerant materials and 1 excellent material were obtained.
