基于ISSR不同繁殖方式半夏的遗传多样性研究

Study on Genetic Diversity of Pinellia ternata With Different Reproduction Methods Based on ISSR

  • 摘要: 以半夏人工种子苗、组培苗和实生苗为试材,采用ISSR分子标记方法,研究了不同繁殖方式半夏的遗传多样性,以期为促进半夏规模化生产提供参考依据。结果表明:从18条引物中筛选出8条多态性较好的ISSR引物,共扩增出条带109条,多态性百分比75%。Nei′s基因多样性范围为0.097 6~0.748 0,大小按顺序排列为人工种子>大田>组培;Shannon多样性指数范围为0.108 1~0.348 2,大小按顺序排列为大田>组培>人工种子;3种繁殖方式之间存在有38.03%的遗传分化;对不同繁殖方式半夏之间的遗传距离分析发现,组培半夏和大田半夏的亲缘关系比人工种子和大田半夏的更接近;聚类分析显示,人工种子半夏和组培半夏聚为一类;大田居群不同叶型遗传多样性分析,芍药叶型和桃叶型半夏亲缘关系最为接近,五叶半夏遗传分化最强。因此,人工种子半夏的基因多样性比组培及大田半夏的基因多样性丰富,半夏人工种子比组培苗更适合用于大田实际生产。

     

    Abstract: Taking Pinellia ternata artificial seed seedlings, tissue culture seedlings and real seedlings as the test materials, ISSR molecular marker method was used to study the genetic diversity of different reproduction methods in Pinellia ternata,in order to provide reference for promoting Pinellia ternata large-scale production.The results showed that 8 ISSR primers with good polymorphisms were screened from 18 primers, and a total of 109 bands were amplified, with a polymorphism percentage of 75%.Nei′s gene diversity ranged from 0.097 6 to 0.748 0,and the sizes were arranged in order for artificial seeds>field>tissue culture.The Shannon diversity index ranged from 0.108 1 to 0.348 2,and the size was sorted as field>tissue culture>artificial seeds.There was a genetic differentiation of 38.03% between the three reproductive methods.The analysis of the genetic distance between different reproductive methods showed that the kinship between tissue culture and field Pirellia terrate was closer than that of artificial seeds and field Pirellia terrate.Cluster analysis showed that artificial seeds and tissue culture were clustered into one category.Analysis of the genetic diversity of different leaf types in the field residential group, among which peony leaf type Pinellia ternata and peach leaf type Pinellia ternata were the closest relatives, five-leaf Pinellia ternata genetic differentiation was the strongest.Therefore, the genetic diversity of artificial seeds in Pinellia ternata was richer than that of tissue culture and field Pinellia ternata,and the Pinellia ternata artificial seed was more suitable for actual production in the field than the tissue culture seedlings.
