金莲花总生物碱分离及其活性研究

Separation and Activity of Total Alkaloids From Trollius chinensis

  • 摘要: 以金莲花为试材,使用响应面法优化总生物碱提取分离工艺,并分析测定金莲花总生物碱抗氧化和抑菌活性,以期为金莲花的深入开发和综合利用提供参考依据。结果表明:最佳提取分离工艺为90%乙醇,液料比39∶1 mL·g-1,超声功率400 W,超声32 min,总生物碱得率为0.844 mg·g-1;对DPPH、ABTS自由基及超氧阴离子清除率的IC50(半抑制浓度)值分别为0.016、0.036、4.404 mg·mL-1;对大肠杆菌和金黄色葡萄球菌的MIC值分别为1.990 mg·mL-1和1.330 mg·mL-1。该研究初步证实金莲花富含生物碱类物质,且具有较强的抗氧化和抑菌活性。

     

    Abstract: The extraction and separation process of total alkaloids from Trollius chinensis Bunge was optimized by response surface methodology.The antioxidant and antibacterial activities of total alkaloids were studied, in order to provide reference for the further development and comprehensive utilization of Trollius chinensis Bunge.The results showed that the best conditions for extraction and separation of total alkaloids were as follow, ethanol concentration 90%,liquid-solid ratio 39∶1 mL·g-1,ultrasonic power 400 W,ultrasonic time 32 minutes, total alkaloids yield was 0.844 mg·g-1.The IC50(half maximal inhibitory concentration) values of total alkaloids on DPPH,ABTS free radical, and superoxide anion scavenging rate were 0.016,0.036 mg·mL-1and 4.404 mg·mL-1,and the MIC values of total alkaloids against Escherichia coli and Staphylococcus aureus were 1.990 mg·mL-1 and 1.330 mg·mL-1.This study preliminarily confirmed that Trollius chinensis Bunge was rich in alkaloids, which had strong antioxidant and antibacterial activities.
