光质和光强对‘蓝鼠耳’玉簪试管苗的影响

Effects of Light Quality and Light Intensity on ‘Blue Mouse Ears’ Hosta Tissue Culture Plantlets

  • 摘要: 以‘蓝鼠耳’玉簪为试材,利用LED光源,研究了不同光质和光强对试管苗生长及生根的影响,以期为‘蓝鼠耳’玉簪试管苗的工厂化生产提供参考依据。结果表明:1)LED光质为W∶R=7∶1时,试管苗的增殖系数为4.71,是所有处理中的最大值;株高和叶面积综合表现良好,分别为17.48 mm和30.90 mm2,说明W∶R=7∶1时对试管苗的生长有较好的促进作用。试管苗的生根率在R∶B=5∶1、W∶R=7∶1、R∶W=2∶3时均高于97%,优于其它处理。LED光质为R∶W=2∶3时试管苗的根数为17.54条,说明光质R∶W=2∶3时能够有效促进试管苗根系生长。2)低光强59μmol·m-2·s-1时有利于试管苗增殖,随着光照强度的增加,增殖系数呈下降趋势。当光强为176~235μmol·m-2·s-1时,试管苗的叶面积表现良好,为73.09 mm2,说明该光强对试管苗的生长有较好的促进作用。当光照强度为118~176μmol·m-2·s-1时试管苗的最大根长为7.44 mm,说明该光强可以促进试管苗根系的生长。最低光强(0~59μmol·m-2·s-1)和最高光强(706~824μmol·m-2·s-1)不利于试管苗的生长。

     

    Abstract: ‘Blue Mouse Ears’ Hosta was used as the research material.The effects of different light quality and light intensity on the growth and rooting of tissue culture plantlets were studied by LED,the suitable light quality and light intensity were selected, in order to provide reference for the industrialized production of ‘Blue Mouse Ears’ Hosta plantlets.The results showed that, 1)when the LED light quality was W∶R=7∶1,the proliferation rate of tissue culture plantlets was 4.71,which was the highest among all treatments; the comprehensive performance of plant height and leaf area was good, which were 17.48 mm and 30.90 mm~2 respectively, indicated that W∶R=7∶1 could promote the growth of tissue culture plantlets.Treatments of R∶B=5∶1,W∶R=7∶1 and R∶W=2∶3 had a rooting rate higher than 97%,which were better than other treatments.When the LED light quality was R∶W=2∶3,the root number of plantlets was 17.54,which indicated that the light quality R∶W=2∶3 could promote the root growth of tissue culture plantlets effectively.2)The lowest light intensity 59 μmol·m-2·s-1 was beneficial to the proliferation of tissue culture plantlets, with the increase of light intensity, the proliferation coefficient decreased.When the light intensity was 176-235 μmol·m-2·s-1,the leaf area of tissue culture plantlets were good, and the numerical value was 73.09 mm~2 respectively, indicated that the light intensity of 176-235 μmol·m-2·s-1 could promote the growth of tissue culture plantlets.When the light intensity was 118-176 μmol·m-2·s-1,the maximum root length was 7.44 mm, which indicated that the light intensity in the range of 118-176 μmol·m-2·s-1 could promote the root growth of tissue culture plantlets.The lowest light intensity(0-59 μmol·m-2·s-1) and the highest light intensity(706-824 μmol·m-2·s-1) were not conducive to the growth of plantlets.
