尖孢镰刀菌中S-腺苷甲硫氨酸依赖性甲基转移酶的功能研究

Functional Study on AdoMet-MTases in Fusarium oxysporum

  • 摘要: 以尖孢镰刀菌粘团专化型(Fusarium oxysporum f.sp.conglutinans,FOC,58385)2号生理小种为试材,采用同源重组和PEG介导原生质体转化的方法对尖孢镰刀菌中含有编码S-腺苷甲硫氨酸依赖性甲基转移酶结构域蛋白的基因进行敲除,通过分析野生型与突变体的表型差异,研究了该蛋白在尖孢镰刀菌中的生物学功能,以期为枯萎病的防治提供参考依据。结果表明:缺失突变体相对于野生型菌株,气生菌丝稀疏、分支少、产孢量和孢子萌发率显著增加,对氧胁迫和渗透胁迫不敏感,接种甘蓝幼苗后致病力显著增强,表明该基因在尖孢镰刀菌中可能与各类胁迫调节相关,通过负调节生长发育过程,维持尖孢镰刀菌的各项稳态。

     

    Abstract: Physiological race 2 of Fusarium oxysporum f.sp.conglutinans was used as experimental material.Homologous recombination and PEG mediated protoplast transformation were used to knock out the gene encoding AdoMet-MTases in F.oxysporum.The physiological function of the protein in F.oxysporum was studied by comparing phenotypic differences between wild type and mutants, in order to provide referance for the control of Fusarium wilt.The results showed that the deletion mutants had fewer hyphae,fewer branches,higher sporulation and spore germination rate than the wild-type strain.The mutants were not sensitive to oxygen stress and osmotic stress,and showed a significant increase in pathogenicity on cabbage seedlings.The results suggested that the gene might be involved in regulation of various stresses and maintained homeostasis through negative regulation of growth and development in F.oxysporum.
