响应面法优化超声辅助提取北柴胡黄酮工艺及抗氧化活性

Optimization of Ultrasonic Assisted Extraction of Flavonoids From Bupleurum chinense by Response Surface Methodology and Its Antioxidant Activity

  • 摘要: 以北柴胡为试材,采用超声辅助法提取总黄酮,通过单因素试验和响应面试验优化提取工艺,研究了总黄酮对DPPH自由基和ABTS自由基的清除能力,以及不同部位柴胡总黄酮的含量,以期为开发利用柴胡提供参考依据。结果表明:试验模型极显著(P<0.000 1),且模型预测值与真实值间拟合度较好(R2=0.972 2)。响应面优化超声辅助提取柴胡总黄酮最佳提取条件为超声温度57℃,超声功率170 W、料液比1∶156 mg·mL-1、乙醇浓度74%,柴胡总黄酮得率8.21%,与预测值(8.17%)吻合较好。柴胡不同部位总黄酮含量顺序为叶>茎>根,且不同部位总黄酮对DPPH自由基和ABTS自由基均具较强的清除作用。

     

    Abstract: Taking Bupleurum chinense as the test material, extraction of total flavonoids from Bupleurum chinense by assisted with ultrasound was studied, the extraction process was optimized through single factor test and response surface test.The clear ability of total flavonoids to DPPH free radicals and ABTS+ free radicals and the content of total flavonoids in different parts of Bupleurum chinense Maxim were studied, in order to provide reference for the development and utilization of Bupleurum chinense.The results showed that the experimental model was very significant(P<0.000 1),and the goodness of fit between the predicted value of the model and the true value was good(R~2=0.972 2).The optimum extraction conditions of ultrasonic assisted extraction of total flavonoids from Bupleurum chinense by response surface methodology were as follows, ultrasonic temperature 57 ℃,ultrasonic power 170 W,solid-liquid ratio 1∶156 mg·mL-1,ethanol concentration 74%.The yield of total flavonoids from Bupleurum chinense was 8.21%,which was in good agreement with the predicted value(8.17%).The order of total flavonoids content in different parts of Bupleurum chinense was leaf>stem>root.Total flavonoids from different parts had strong scavenging effects on DPPH free radicals and ABTS+ free radicals.
