AtWUS过表达诱导分蘖洋葱体细胞胚高频发生

AtWUS Promotes High Frequency Somatic Embryogenesis Potato Onion (Allium cepa L. var. Aggregatum G. Don)

  • 摘要: 以分蘖洋葱茎尖外植体诱导的愈伤组织为试材,采用农杆菌介导法,将外源基因AtWUS导入分蘖洋葱愈伤组织中,研究AtWUS对分蘖洋葱愈伤组织诱导体细胞胚发生的影响,以期建立分蘖洋葱体细胞胚高频发生体系。结果表明:过表达AtWUS可以显著促进分蘖洋葱愈伤组织的体细胞胚发生率(大约6倍),91%的双极性体细胞胚具有发育形成完整植株能力。RT-PCR分析结果表明,AtWUS在体细胞胚和转基因植株中高丰度表达。

     

    Abstract: Taking callus induced from shoot apex explants of potato onion(Allium cepa var. aggregatum Don) as test material, the foreign gene AtWUS genetic transformation was carried out by the agrobacterium-mediated transformation method with potato onion callus.The effects of AtWUS on somatic embryogenesis(SE) of potato onion calluswas studied, in order to develop a high-frequency SE regeneration systemvia over-expressing AtWUS from in potato onion callus.The results showed that overexpression of the AtWUS gene incallus promoted, as the percentage of explants giving rise to embryogenic tissues was significantly higher(×6) when AtWUS gene was over-expressed than in the control.Bipolar somatic embryos resulted 91% complete plant regeneration.RT-PCR analysis showed that AtWUS gene was highly expressed in somatic embryos and transgenic plants.
