Abstract:
Taking the branches and leaves of Taxus media as experiment materials, on the basis of single factor experiment, the response surface method was used to optimize the thermal reflux ectraction process of total flavonoid from Taxus media,and the scavenging rate of DPPH·,ABTS
+· and nitrite radicals were used to study the antioxidant activity of total flavonoid, in order to provide a reference for the further development and utilization of T.media.The results showed that the optimal extraction process was as following, the liquid-material ratio was 32∶1 mL·g
-1,ehanol concentration was 63%,extraction temperature was 71 ℃ and extraction time was 86 minutes, the number of extraction was 2 times.Under the above conditions, the extraction yield of total flavonoid from T.media was up to 128.10 mg·g
-1.The extraction rate of total flavonoid was higher in extraction process, therefore the extraction technology of total flavonoid from T.media was stable and applicable.At the maximum total flavonoid extract concentration, the scavenging rates of DPPH·,ABTS
+· and nitrite were 91.04%,99.17% and 65.50%,respectively.The result indicated that the antioxidant activity from T.media was strong in vitro.
