Abstract:
The D. odorifera T. Chen of nodules were used as experimental materials under the cultivation of three different gravel and loam ratios,and the endophytic rhizobia were isolated and purified.They were identified by morphological observation,biological characteristics analysis and 16SrDNA sequencing.The species difference and biological characteristics of endophytic rhizobia from Dalbergia odorifera T. Chen under different gravel content culture substrate were studied,in order to provide candidate strains for popularization and cultivation of D. odorifera T. Chen in rocky desertification areas.The results showed that Rhizobium and Bradyrhizobium were the strains in whole gravel culture substrate,while strains in gravel and loam mixed substrate and whole loam substrate were Rhizobium.The rhizobia isolated in this study had strong nitrogen-fixing nodules and high concentration calcium tolerance ability.The main root length,fresh weight and dry weight of D. odorifera T. Chen seedlings were significantly increased(P<0.05).The species of endophytic rhizobia of D. odorifera T. Chen with different gravel content were different and all of them could nodulate and promote plant growth,which indicated that these rhizobia had potential application prospect in rocky desertification soil environment with different gravel contents.
