适于新疆干旱地区黑木耳液体菌种培养条件优化研究

Study on Liquid Fungus of Auricularia auricula Suitable for Southern Xinjiang and Optimization of Its Culture Conditions

  • 摘要: 以8个黑木耳(Auricularia auricula)菌种为试材,通过观察固体菌种(不同pH)和液体菌种菌丝生长状况和时间,从PDA培养基、麸皮培养基、玉米粉培养基和PDA改良培养基4种培养基中筛选出最优培养基,以期对优质菌种在最优培养基的培养条件进行优化。结果表明:PDA固体培养基中,XLG-1、XLG-2、XLG-5和XLG-6的生长速度快、污染率低、菌丝生长势强,其中,XLG-1和XLG-2在pH分别为6和9的PDA固体培养基中生长速度显著高于其它菌种,在PDA改良培养基上的菌丝体生物量显著高于其它培养基,且二者菌丝体生物量之间差异不显著,XLG-1的菌丝体密度大于XLG-2、直径小于XLG-2。二者在PDA改良培养基中的最佳培养条件为装量100 mL,温度25℃,接种量10%。综上,XLG-1和XLG-2为适应性强、生长速度快、生物量大菌株,其中XLG-2属于耐高pH的优质菌株,适合南疆地区栽培,PDA改良培养基是最佳培养基。

     

    Abstract: Eight A.auricula strains as materials, the optimal medium is selected from PDA medium, bran medium, corn flour medium and PDA improved medium based on the mycelial growth status, growth time and the biomass of solid strains(different pH) and liquid strains, and the culture conditions of high-quality strains in the optimal medium were optimized.The results showed that the mycelium of XLG-1,XLG-2,XLG-5 and XLG-6 grew faster and they had lower pollution rate and stronger mycelial growth potential than others, in PDA solid medium.Among the mycelium of XLG-1,XLG-2,XLG-5 and XLG-6,XLG-1 and XLG-2 grew faster than others with pH 6 and 9,respectively, in PDA solid medium.The mycelial biomass of XLG-1 and XLG-2 from PDA improved medium was significantly higher than that from other media.The optimum culture conditions of XLG-1 and XLG-2 in PDA improved medium were as follows, 100 mL of medium loading, 25 ℃ of temperature and 10% of inoculation amount.In conclusion, XLG-1 and XLG-2 had strong adaptability, fast growth rate and large biomass.PDA improved medium was the best medium.XLG-2 was a high-quality strain with high pH resistance and was suitable for cultivation in Southern Xinjiang.
