Abstract:
Taking the young stem segments with axillary buds of a new variety of Lagerstroemia indica ‘Wuye’ as explants, the orthogonal experimental method was used to study the effects of disinfection time of different combinations of NaClO and C
2H
5OH on the contamination and germination of the explants.The effects of three basic media, WPM,1/2MS,and MS,as well as different concentration combinations of 6-BA and NAA,on the induction and proliferation of axillary buds from tender stems of L.indica ‘Wuye’ were studied, as well as the effects of three basic media and different concentration combinations of IBA on the rooting of L.indica ‘Wuye’ were studied, in order to provide reference for mass propagation of Lagerstroemia indica ‘Wuye’.The results showed that, when the disinfection time of 75% C
2H
5OH was 30 seconds and the disinfection time of 5% NaClO was 6 minutes, the pollution rate was lower, at 18.6%.The WPM medium was suitable for the induction and proliferation of axillary buds in stem segments of L.indica ‘Wuye’.The highest induction rate was achieved when the concentrations of 6-BA and NAA were 1.0 mg·L
-1 and 0.5 mg·L
-1,respectively, reaching 93.28%.When the concentration of 6-BA was 1.0 mg·L
-1 and the concentration of NAA was 0.1 mg·L
-1,the highest proliferation coefficient was 5.83.The concentration change of IBA had a significant impact on the rooting of L.indica ‘Wuye’.Adding 0.5 mg·L
-1 IBA to the 1/2MS had the highest rooting rate of 92.99%,and the growth of sterile seedlings was good.After refining the seedlings, transplanted the tissue culture seedlings onto a substrate of peat soil∶perlite∶vermiculite in a ratio of 3∶1∶2,with a survival rate of up to 91%.
