基于响应面法优化冬荪液体菌种培养基

Optimizing the Liquid Culture Medium of Phallus dongsun by Response Surface Methodology

  • 摘要: 以冬荪菌株“PD1710141”为试材,采用Plackett-Burman设计、最陡爬坡试验、Box-Benhnken设计响应面法研究了冬荪液体菌种培养基的优化,得到成本低、菌丝生物量高的冬荪液体菌种培养基配方,以期为工厂化大规模生产冬荪栽培种提供参考依据。结果表明:当摇瓶转速150 r·min-1、装液量150 mL时,冬荪液体菌种最佳配方为葡萄糖10 g·L-1、磷酸二氢钾1 g·L-1、蛋白胨2.5 g·L-1、硫酸镁0.8 g·L-1、马铃薯380 g·L-1、酵母粉1.5 g·L-1、维生素B1 0.03 mg·L-1,其冬荪菌丝鲜质量可达(4.300±0.038)g、为模型预测值的97%。综上可知,响应面法可应用于冬荪液体培养基配方及条件优化。

     

    Abstract: Taking the strain ‘PD1710141’about Phallus dongsun as the test material,the response surface method were used to optimize the liquid culture medium,which involved Plackett-Burman design,path of steepest ascent and Box-Benhnken design,get the liquid culture medium with low cost and high yield,in order to provide reference for mass production of cultivated species about Phallus dongsun.The results showed that when the rotating speed was 150r·min-1 and the liquid volume of shake flask was 150mL,the composition of optimal liquid culture medium about Phallus dongsun were 10g·L-1 glucose,1g·L-1 potassium dihydrogen phosphate,2.5g·L-1 peptone,0.8g·L-1 magnesium sulfate,380g·L-1 potato,1.5g·L-1 yeast extract powder and 0.03mg·L-1 vamin B1.Implementing this optimal medium,the fresh weight of mycelia was(4.300±0.038)g and was 97% of the predicted value.In conclusion,response surface method could be applied to the optimization of liquid culture medium and condition.
