Abstract:
Taking the strain ‘PD1710141’about Phallus dongsun as the test material,the response surface method were used to optimize the liquid culture medium,which involved Plackett-Burman design,path of steepest ascent and Box-Benhnken design,get the liquid culture medium with low cost and high yield,in order to provide reference for mass production of cultivated species about Phallus dongsun.The results showed that when the rotating speed was 150r·min-1 and the liquid volume of shake flask was 150mL,the composition of optimal liquid culture medium about Phallus dongsun were 10g·L
-1 glucose,1g·L
-1 potassium dihydrogen phosphate,2.5g·L
-1 peptone,0.8g·L
-1 magnesium sulfate,380g·L
-1 potato,1.5g·L
-1 yeast extract powder and 0.03mg·L
-1 vamin B1.Implementing this optimal medium,the fresh weight of mycelia was(4.300±0.038)g and was 97% of the predicted value.In conclusion,response surface method could be applied to the optimization of liquid culture medium and condition.
