响应面法优化平菇产漆酶的培养条件

Optimization of Culture Conditions for Laccase Production From Pleurotus ostreatus by Response Surface Methodology

  • 摘要: 以平菇680为试材,以漆酶为响应值,在单因素试验的基础上,采用响应面Box-Behnken设计建立数学模型,对平菇液体培养基进行了优化,以期为实现木质纤维的快速降解提供参考依据。结果表明:葡萄糖、酵母膏、铜离子及接种量为影响漆酶活性的主要因子,最佳液体培养基为葡萄糖52g·L-1、酵母膏9g·L-1、铜离子1.5mmol·L-1、接种量3%,在该优化培养基条件下得到漆酶活性为359.12U·g-1,与预测值358.77U·g-1相近,说明响应面设计对于优化液体培养基以提高漆酶活性切实可行。

     

    Abstract: Pleurotus ostreatus was used as the test material.On the base of single factor experiment,Box-Behnken design in response surface methodology were employed to establish mathematical model for optimization of liquid medium composition of Pleurotus ostreatus while the laccase activity was taken as the response in this study.In order to provide a theoretical basis and reference for the rapid degradation of lignin fiber.The results showed that glucose,yeast paste,Cu2+ and inoculation size were main composition for the laccase activity,the optimal composition to produce laccase from Pleurotus ostreatus was as follows glucose 52 g·L-1,yeast paste 9 g·L-1,Cu2+1.5 mmol·L-1,inoculation size 3%.Under the optimized conditions,the laccase activity was 359.12 U·g-1,which agreed closely with the theoretical result of 358.77 U·g-1 predicted by the developed model.Above results indicate that the response surface methodology was viable for optimizing medium composition in improving laccase activity.
