Abstract:
Four species of Phellinus,Sanghuangporus vaninii JS1,Sanghuangporus vaninii LS2,Inonotus vaninii YS3 and Phellinus igniarius S6 were optimized through the optimization of Sanghuangporus parent species, original species and fruiting bodies cultivation.The Sanghuangporus lindenus mycelial growth and fruiting bodies active components were analysed for these four species.The content of total triterpenes, flavonoids and polysaccharides extracts in fruiting bodies were compared by ultraviolet colorimetric colorimetry, in order to provide reference for the study of functiomal active substances and the quality control of fruiting bodies.The results showed that there were significant difference in mycelial germination rates at different stages were observed.The growing of four Sanghuangporus species on PDA-enriched medium with sucrose as carbon source were the fastest.The growth was relatively slower on the corn meal and bran medium, and on the mulberry branch filtrate medium was the slowest.The growth phenotype of Inonotus vaninii YS3 mycelium was the best in terms of growth on the mulberry branch filtrate medium.The growth of Sanghuangporus philoxeroides were the fastest after the third day of culture, JS1,YS3 and S6 mycelial grew the fastly in bran medium, and the growth rates were 4.78 mm·d
-1,4.60 mm·d
-1 and 3.08 mm·d
-1,respectively.There was no significant difference in the growth rate of LS2 in the four media, among which the fastest was 4.17 mm·d
-1 in the corn meal medium.The content of polysaccharides in the fruit body of LS2 was the highest, the total flavonoids and triterpenes in YS3 fruiting bodies were the highest, and the contents of flavonoids, triterpenes and polysaccharides in S6 fruiting bodies were all lowest. Inonotus vaninii YS3 and Sanghuangporus vaninii LS2 could be used as excellent production strains considering the growth rate of mycelial, agronomic characters and active component content of fruiting bodies.
