响应面优化三叶木通总黄酮的提取工艺及其抗氧化活性

Optimization of Extraction Conditions of Flavonoids From Akebia trifoliata by Response Surface Methodology and Its Antioxidant Activity

  • 摘要: 以三叶木通为试材,采用单因素试验结合响应面法优化三叶木通提取总黄酮的工艺,研究了总黄酮提取物的抗氧化活性,以期为三叶木通的进一步开发利用提供参考依据。结果表明:三叶木通总黄酮最佳提取工艺条件为乙醇体积分数56%、提取时长70 min、提取温度90℃、液料比50∶1 mL·g-1,该条件下提取总黄酮的量为60.74 mg·g-1,与预测值60.492 4 mg·g-1基本一致,表明该提取工艺稳定可行。在最佳工艺条件下得到的三叶木通总黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)自由基有较高的清除力,说明三叶木通总黄酮拥有较好的抗氧化活性。

     

    Abstract: Taking Akebia trifoliata as test material, the extraction technology of total flavonoids from Akebia trifoliata was optimized by single factor test combined with response surface method, in order to provide reference for the further development and utilization of Akebia trifoliata.The results showed that the optimal extraction conditions were 56% ethanol volume fraction, 70 minutes extraction time, 90 ℃ extraction temperature and 50∶1 mL·g-1 liquid-solid ratio.Under these conditions, the amount of flavonoids extracted was 60.74 mg·g-1,which was basically consistent with the predicted value of 60.492 4 mg·g-1,indicating that the extraction process was stable and feasible.The total flavonoids obtained under the optimal process conditions had a high scavenging ability on 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH) free radical, indicating that the total flavonoids of Akebia trifoliata had a good antioxidant activity.
