香菇继代培养对菌丝特性和显微结构的影响

摘要: 以香菇菌株"辽抚4号"为试材,采用继代培养的方法,研究了继代培养次数对菌丝的生长特性和显微形态变化的影响,以期为菌种使用和继代培养对菌种退化的研究提供参考依据。结果表明:继代培养对菌丝的生长特性和菌丝微观形态影响显著,子实体组织分离的部位和培养基成分对继代培养菌株菌丝的长速和长势影响明显。随着继代培养次数的增加菌丝长速下降趋势显著,继代培养40次之后,木屑培养基上,菌盖部位分离菌丝生长速度下降35.62%,PDA培养基上,菌盖与菌柄交界处的组织分离菌丝生长速度下降34.05%,菌丝稀疏,长势变弱;菌丝显微结构可见明显变化,菌丝宽度、锁状联合的宽度和数量发生菌丝微观形态的改变,趋势上随着继代培养次数的增加,菌丝分枝和锁状联合数量逐渐减少,菌盖处组织分离得到的菌丝锁状联合的宽度逐渐增加;组织分离菌株的菌丝特性差异表明菌盖、菌盖与菌柄交界处是最佳分离部位。Abstract: The Lentinula edodes strain ’Liaofu-4’ was used as test material, the effects of subculture times on mycelial growth characteristics and micromorphological changes were studied by subculture, in order to provide a reference for strain application and the study of the strain degradation in the process of subculture.The results showed that subculture times had a significant effect on the growth characteristics and micromorphology of mycelia, and the isolate parts of the fruiting body and the composition of culture medium affect the growth rate and state of the subcultured mycelia apparently.With the increase of subculture times, the growth rate of mycelia decreased significantly.After subcultured for 40 times, the growth rate of mycelia isolated from the pileus decreased by 35.62% on sawdust medium, and the mycelia isolated from the junction of the pileus and the stipe decreased by 34.05% on PDA,and the mycelial growth sparse.With the increase of subculture times, the number of mycelia branches and clamp-connection decreased gradually, and the width of mycelia clamp-connection isolated from pileus increased gradually.In conclusion, the subculture of L.edodes could cause the decrease of mycelial growth rate, the weakening of mycelial growth state, and the change of micromorphology including the width of mycelia, the width and quantity of clamp-connection.The study of mycelial characteristics showed isolation part from pileus and the junction of pileus and stalk were better choice.
