怀菊快繁体系及组培苗分级标准的建立

Establishment of Grading Standard of Tissue Culture Seedling of Chrysanthemum morifolium

  • 摘要: 以怀菊叶片为试材,采用正交设计方法,研究了怀菊的快繁体系,以期在继代培养后,确定株高和地茎为最终分级的量化指标。结果表明:最佳灭菌方案为0.1%升汞处理3 min;不定芽诱导的最佳培养基为MS+2.0 mg·L-1 6-BA+0.1 mg·L-1NAA+20 g·L-1蔗糖,其诱导率达到88%;最佳生根培养基为1/2MS+0.1 mg·L-1 IBA,其生根率为100%;通过聚类分析将怀菊组培苗分为Ⅰ级、Ⅱ级和Ⅲ级苗,Ⅰ级组培苗的等级标准为株高≥5 cm、地茎≥1.16 mm,共23株;Ⅱ级组培苗为4 cm≤株高<5 cm、1 mm≤地茎<1.16 mm,共40株;Ⅲ级组培苗为1 cm≤株高<4 cm、地茎<1 mm,共17株,符合正态分布。该研究为怀菊规范化种植以及种苗制定通用标准时提供参考依据。

     

    Abstract: Taking Chrysanthemum morifolium leaves as the test material, the rapid propagation system of Chrysanthemum morifolium was stuided by orthogonal design method.After subculture, the plant height and ground stem were determined as quantitative indicators for the final classfication.The results showed that the best sterilization plan was 0.1% mercury liter treatment for 3 minutes, in order to provide a reference for the standardized planting of Chrysanthemum morifolium and the formulation of general standards for seedlings.The best medium for adventitious bud induction was MS+2.0 mg·L-1 6-BA+0.1 mg·L-1 NAA+20 g·L-1 sucrose, its induction rate reached 88%.The best rooting medium was 1/2 MS+0.1 mg·L-1 IBA,and its rooting rate was 100%.Through cluster analysis, Chrysanthemum morifolium tissue cultured seedlings were divided into Level Ⅰ,Level Ⅱ and Level Ⅲ,the grade standard for Grade I tissue culture seedlings were plant height≥5 cm, ground stem≥1.16 mm, a total of 23 plants.Grade Ⅱ tissue culture seedlings were 4 cm≤plant height<5 cm, 1 mm≤ground stem<1.16 mm, a total of 40 plants.Level Ⅲ tissue culture seedlings were 1 cm≤plant height<4 cm, ground stem<1 mm, a total of 17 plants, in line with the normal distribution.
