大球盖菇液体发酵培养基优化及其胞外多糖抗氧化活性

Optimizing Culture Medium and Antioxidant Activity of Extracellular Polysaccharides in Liquid Fermentation of Stropharia rugosoannulata

  • 摘要: 以大球盖菇D5为试材,采用单因素试验,通过Plackett-Burman设计及响应面法优化大球盖菇的液体发酵培养基,评价大球盖菇胞外多糖的抗氧化活性,以期为大球盖菇液体菌种的生产及其胞外活性多糖的开发提供参考依据。结果表明:最佳液体发酵条件为葡萄糖浓度30.3 g·L-1、酵母膏浓度为3.7 g·L-1、蛋白胨浓度1.0 g·L-1、KH2PO4浓度2.0 g·L-1、MgSO4·7H2O浓度1.0 g·L-1、维生素B1浓度19.0 mg·L-1,此时菌丝生物量和胞外多糖含量分别为1.317 g·(200mL)-1和230.09 mg·L-1,较优化前分别提高37.7%和15.9%,胞外多糖对羟基自由基、DPPH自由基、ABTS+自由基有一定的清除作用,且其自由基的清除率随浓度增加而增大。

     

    Abstract: The liquid fermentation conditions for Stropharia rugosoannulata D5liquid medium in shake flasks were optimized by single factor experiment combined with response surface method,and the antioxidant activity of extracelluar polysaccharides was studied,in order to provide a reference for the production of liquid inoculum and the development of extracellular polysaccharides.The results showed that the optimal liquid fermentation conditions were glucose 30.3 g·L-1,yeast extract3.7g·L-1,peptone 1.0g·L-1,KH2PO42.0g·L-1,MgSO4·7H2O 1.0g·L-1 and vitamin B1 19.0mg·L-1.The yield of mycelia and extracellular polysaccharides under this fermentation process was 1.317g·(200mL)-1 and 230.09 mg·L-1,respectively,which were 37.7%and 15.9%higher than those before optimization.Extracellular polysaccharides was obtained from the fermentation exhibited a strong scavenging effect on Hydroxyl,DPPH and ABTS+free radicals,and the scavenging rate increased with the increase of concentration.
