不同碳源培养条件下黑木耳菌丝代谢组分析

Metabolic Analysis of Auricularia auricula (L.ex Hook.) Underw Mycelium Under Different Carbon Source Culture Conditions

  • 摘要: 以不用碳源培养条件下的黑木耳菌丝为试材,采用液相色谱-质谱联用仪(LC-MS)技术对黑木耳菌丝进行非靶向代谢组学分析,研究了不同碳源处理对黑木耳菌丝代谢物的影响,以期为黑木耳菌丝降解碳源机理提供参考依据。结果表明:基于主成分分析(PCA),样本间差异明显;基于偏最小二乘判别分析结果共鉴定出588个代谢物;通过对显著代谢物进行筛选(P<0.05)共得到66个差异代谢物,主要包括氨基酸及其衍生物、不饱和脂肪酸、有机酸及其衍生物、有机化合物、酚类、脂类、萜类等多种化合物。基于KEGG通路分析,将差异代谢物注释到21个代谢通路,其中7条显著富集的通路(P<0.05)。

     

    Abstract: Taking Auricularia auricula(L.ex Hook.) Underw mycelium cultured without carbon source as test material, the nontargeted metabonomic analysis of Auricularia auricula(L.ex Hook.) Underw mycelium was conducted by liquid chromatography mass spectrometry(LC-MS) technology.The effects of different carbon source treatments on the metabolites of Auricularia auricula(L.ex Hook.) Underw mycelium were studied, in order to provide reference for the mechanism of Auricularia auricula(L.ex Hook.) Underw mycelium degradation of carbon source.The results showed that based on principal component analysis(PCA),there were significant differences between samples; a total of 588 metabolites were identified based on the results of partial least squares discriminant analysis.A total of 66differential metabolites were obtained by screening significant metabolites(P<0.05),including amino acids and their derivatives,unsaturated fat acids,organic acids and their derivatives,organic compounds,phenols,lipids,terpenes and other compounds.Based on KEGG pathway analysis,differential metabolites were annotated into 21metabolic pathways,of which7were significantly enriched(P<0.05).
