Abstract:
The tubers of Pinellia ternata were used as experimental materials.The effects of different types and concentrations of plant growth regulators on adventitious bud induction, proliferation, rooting and the effect of current stimulation on adventitious bud proliferation were studied by tissue culture, in order to provide reference for the establishment of asexual rapid propagation system for Pinellia ternata.The results showed that the optimal medium for adventitious bud induction was MS+1.5 mg·L
-1 6-BA+0.1 mg·L
-1 NAA.The average number of adventitious buds per explant was 9.38 and the induction rate was 93.75%.The use of current stimulation not only increased the proliferation of adventitious buds, but also significantly promoted the growth of Pinellia ternata plants.Especially under the treatment of 20 mA and 5 minutes, the maximum proliferation of adventitious buds was 13.31 times, and the tissue culture seedlings grew robust and rooted.The optimum induction medium for rooting was 1/2 MS+0.2 mg·L
-1 NAA,the rooting rate was 100%,and the number of rooting was 23.75.Asexual rapid propagation system of Pinellia ternata was established to provide reference for high-quality seedling breeding and factory production.
