不同栽培配方对茶树菇生长发育及胞外酶活性的研究

Effects of Different Cultivation Formulas on Growth and Extracellular Enzyme Activity of Cultured Agrocybe cylindracea

  • 摘要: 以五节芒和棉籽壳为主要原料栽培茶树菇,按照不同添加比例设计栽培配方,研究了5种熟料栽培配方对茶树菇菌丝生长状况、生物学效率、农艺性状和5种胞外酶活性影响的规律,以期为提高五节芒等菌草资源的综合利用价值及促进我国菌业可持续发展提供参考依据。结果表明:P3配方对茶树菇菌丝长势、农艺性状、第一潮菇产量和生物学效率均表现最佳,均优于其它配方,其菌丝长势整齐浓密,出菇时间最短,子实体形态特征最优,头潮菇产量为(317.187±1.980)g·袋-1,生物学效率为63.44%;不同栽培配方下羧甲基纤维素酶、木聚糖酶、淀粉酶、漆酶和蛋白酶等胞外酶活性在茶树菇整个生育期内的活性变化具有明显的阶段性,且同一种酶在不同的栽培料配方中活性变化趋势基本一致;通过菌丝生长速度与5种胞外酶的相关性分析,发现羧甲基纤维素酶、木聚糖酶和淀粉酶与菌丝生长速度均呈正相关,其中木聚糖酶和淀粉酶呈极显著正相关,说明菌丝生长速度快的栽培配方能促进菌丝胞外酶的分泌,为了保障营养的供应,菌丝体分泌更多的羧甲基纤维素酶、木聚糖酶和淀粉酶对栽培料进行降解。综合分析,P3配方最适宜茶树菇生长。

     

    Abstract: Agrocybe cylindrace were cultivated with Miscanthus floridulus and cottonseed shells as the main raw materials, and design of cultivation formulations according to different addition ratios, the effects of five clinker cultivation formulations on mycelium growth, biological efficiency, agronomic traits and five extracellular enzyme activities of A.cylindracea were studied, in order to provide reference for improving the comprehensive utilization value of mushroom resources such as Miscanthus floridulus and promoting the sustainable development of China′s mushroom industry.The results showed that formulation P3 had the best performance on mycelial growth, agronomic characteristics, first crop yield and biological efficiency of A.cylindracea,these traits were superior to other formulations, with neat and dense mycelial growth, the shortest germination time, the best morphological characteristics of fruiting entities, the first tide mushroom yield of(317.187±1.980) g per bag, and the biological efficiency of 63.44%.The activity changed of extracellular enzymes such as CMCase, xylanase, amylase, laccase and proteinase in the whole growth period of A.cylindracea under different cultivation formulas had obvious stages, and the trend of activity of the same enzyme in different cultivar formulations was generally consistent.The correlation analysis between mycelial growth rate and five extracellular enzymes showed that CMCase, xylanase and amylase were positively correlated with mycelial growth rate, among which xylanase and amylase were highly significantly positively correlated, indicating that the cultivation formula with fast mycelial growth rate could promote the secretion of mycelial extracellular enzymes, and in order to guarantee the supply of nutrients, the mycelium secreted more CMCase, xylanase and amylase to degrade the cultivated material.Comprehensive analysis, the P3 formulation was most suitable for the growth of tea tree mushrooms.
