百合斑驳病毒靖宇分离物全基因组序列测定与进化分析

Sequencing and Analysis of the Genome of Lily mottle virus (LMoV) Jingyu Isolate

  • 摘要: 以采自吉林省白山市靖宇县的野生百合感病叶片为试材,采用小RNA深度测序法检测到1株百合斑驳病毒(Lily mottle virus,LMoV),采用分段克隆方法,对LMoV靖宇分离物(LMoV-JY)的全基因组进行测定并分析,以期对吉林省百合病毒病的检测和防控提供参考依据。结果表明:LMoV-JY基因组大小为9 648个核苷酸(nt)序列(GenBank登录号MT795719),该核苷酸序列在第154~9 444位存在1个大的开放阅读框(ORF),编码1个多聚蛋白(分子量351.46 kD)。LMoV-JY与GenBank中登录的其他LMoV分离物的全基因组核苷酸序列比较,其一致性为82.82%~97.85%,在氨基酸水平上的一致性为92.96%~98.64%,其中与百合斑驳病毒大连分离物LMoV-DL(HM222521)的一致性在该2种水平上均为最高。对比LMoV-JY外壳蛋白与其他54个分离物的氨基酸序列,发现可将LMoV分离物分为2个类群。

     

    Abstract: The infected leaves of wild lily were taken from Jingyu county, Baishan city, Jilin Province.Lily mottle virus(LMoV) was detected by small RNA deep sequencing and the whole genome of LMoV Baishan-Jingyu isolate(LMOV-JY) was determined and analyzed by segmentation cloning method, in order to provide reference for detection, prevention and control of lily virus disease in Jilin Province.The results showed that the genome of LMoV-JY consisted of 9 648 nucleotides(nt)(Genbank Accession No.MT795719).A large open reading frame(ORF) located at nt positions from 154 to 9 444 which encoding a polypeptide of 351.46 kD.By compairing with other isolates of LMoV,the nucleotide identity was from 82.82% to 97.85%,and the amino acid identity was from 92.96% to 98.64%,and it shared the highest identity with the Dalian isolate LMoV-DL(HM222521).Amino acid sequences comparison of the CP gene of LMoV-JY with other 54 isolates in GenBank showed that all of the isolates could be divided into two subgroups.
