Establishment of Transient Gene Overexpression System in Dianthus chinensis

  • D.chinensis was used as test materials.The vector pBI121-GUS was transformed into Agrobacterium tumefaciens GV3101 and EHA105,respectively.After the detached flower buds of D.chinensis infected, the flower bud blooming rate and GUS staining were studied, in order to provide reference for the rapid screening of genes aftecting the expression of flower color.The results showed that after infection with Agrobacterium GV3101,the flower bud blooming rate was 56.67%,which was 15.84 % higher than that of EHA105.The color of GUS staining was obvious when the OD600 of the infiltration solution was 1.4±0.2.In conclusion, Agrobacterium GV3101 with its OD600 of 1.4±0.2 should be better to be used in the transient overexpression system of D.chinensis.
