Molecular Identification and Phylogenetic Analysis of Three Wild Edible Mushrooms in Qinhuangdao Area

Abstract
In order to lay a foundation for the rational development and artificial domestication of wild rou mushroom,ga mushroom and song mushroom in Qinhuangdao area,the ITS regions of rDNA genes were amplified using ITS1 and TIS4 as primers,cloned and sequenced.ITS sequences were analyzed via NCBI blast program,meanwhile phylogenetic tree were constructed,respectively,in order to provide a foundation for the rational development and artificial domestication of the three wild edible mushrooms.The results showed that rou mushroom had 99% similarity with Chroogompus rutilus(MG457851.1),the similarity between ga mushroom and Lactarius akahatsu(EF685045.1)was up to 100%,but song mushroom merely had 96% similarity with Suillus granulatus(MF421107.1).Phylogenetic analysis showed that there was a close genetic relationship between rou mushroom and C.rutilus,their genetic distance reached to 0.Ga mushroom and L.akahatsualso had higher relationship,the genetic distance between them was 0.Song mushroom had a lower genetic relationship to Suillus granulatus(0.033).Therefore,rou mushroom and ga mushroom in Qinhuangdao area were at the molecular level identified as C.rutilus and L.akahatsu,respectively.Song mushroom,which was another kind of wild edible fungi in Qinhuangdao area,belonged to Suillus.
