液态发酵鸡枞菌的条件优化

Optimization of Liquid Fermentation Conditions for Collybia albuminosa

  • 摘要: 鸡枞菌生物多糖含量较高,对人体具有抗肿瘤和增强机体免疫力等多种生理功能,但由于自身生长条件要求较高和生态环境的变化,导致采用传统方法培养的鸡枞菌生物多糖含量较低。通过测定鸡枞菌生物多糖含量、菌体单位体积湿重等指标,在单因素试验的基础上,以多糖含量为考察指标,采用响应面法优化培养时间、培养温度、初始pH值,研究鸡枞菌生物多糖含量达到最大值的最佳培养条件。结果表明,最佳培养条件为:葡萄糖添加量2.50%,蛋白胨添加量0.25%,培养时间7 d,培养温度27.6℃,初始pH为7.0,此优化条件下得到的生物多糖含量达2.71 g/L。该研究为液态培养鸡枞菌提高生物多糖含量提供了理论依据。

     

    Abstract: Collybia albuminosa has higher levels of biological polysaccharide that has various physiological functions such as anti-tumor and enhancement of body immunity, but due to the high requiments of its own growth conditions and the change of ecological environment, the content of Collybia albuminosa biological polysaccharide cultured by traditional method is lower. On the basis of single factor experiments in biological polysaccharide content and bacterial wet weight per unit volume, the best culture conditions including cultivation time,temperature, initial p H value were optimized by response surface method with Collybia albuminosa biological polysaccharide content as index. The results showed that the best culture conditions were as follows: glucose2.50%, peptone 0.25%, culture time 7 days, culture temperature 27.6 ℃, and initial pH 7.0. The content of polysaccharides reached 2.71 g/L under the above conditions. Which provides theoretical basis for the improvement of biological polysaccharide content by liquid culture of Collybia albuminosa.
