西瓜PME基因生物信息学分析及克隆

Bioinformatics analysis and cloning of watermelon PME gene

  • 摘要: 研究从拟南芥(Arabidopsis thaliana)4个果胶甲酯酶基因和2个来自东北农业大学西甜瓜分子育种课题组西瓜转录组数据分析的果胶甲酯酶基因入手,通过BLAST比对方法在葫芦科数据库中鉴定甜瓜(Cucumis melo L.)、西瓜(Citrullus lanatus)果胶甲酯酶同源基因,预测分析其生物信息学。结果表明,各级结构及理化性质与拟南芥果胶甲酯酶基因较为相似,并对其构建系统进化树,建树结果表明,葫芦科各物种间果胶甲酯酶基因家族同源性较高。此外,利用同源克隆方法获得Cla012554基因全长并分析该基因时空表达量,测序结果显示,克隆的Cla012554基因序列与比对序列间总体相似度为95.62%。实时荧光定量PCR分析表明,Cla012554基因在西瓜根、茎、叶和果肉中均表达,其中,茎表达量最高,根表达量最低。同时,Cla012554基因还可能参与西瓜果肉成熟软化。研究为进一步探究葫芦科作物PME基因功能提供重要参考。

     

    Abstract: This study started with four pectin methylesterase genes from Arabidopsis thaliana and two pectin methylesterase genes from the watermelon transcriptome data analysis of the West Muskmelon Molecular Breeding Research Center of Northeast Agricultural University. The method was used to identify the pectin methylesterase homologous genes of melon(Cucumis melo L.) and watermelon(Citrullus lanatus) in the Cucurbitaceae database, and perform their bioinformatics prediction analysis. The results showed that its structure and physicochemical properties were similar to those of Arabidopsis pectin methylesterase genes, and a phylogenetic tree was constructed for them.The tree results showed that the pectin methylesterase gene families of various species were different.The homology was high. In addition, this experiment also used the homologous cloning method to obtain the full-length of the watermelon Cla012554 gene and analyze the temporal and spatial expression of the gene. The sequencing results showed that the overall similarity between the cloned watermelon Cla012554 gene sequence and the comparison sequence was 95.62%, real-time fluorescent quantitative PCR analysis showed that Cla012554 gene was expressed in watermelon roots, stems,leaves and pulp, with the highest expression in the stem and the lowest expression in the root. At the same time, Cla012554 gene might also be involved in the ripening and softening of watermelon pulp.This study provided an important reference for further exploring the functions of PME genes in Cucurbitaceous crops.
