响应面法优化玉米醇溶蛋白提取工艺及抗氧化活性评价
Optimization of zein extraction process by response surface methodology and evaluation of antioxidant activity

摘要: 为制备高品质玉米醇溶蛋白,以玉米蛋白粉为原料,利用超声波辅助提取,优化乙醇提取玉米醇溶蛋白工艺,并评价制备的醇溶蛋白自由基吸附能力。通过单因素和响应面法试验设计优化提取工艺条件。结果表明,乙醇浓度80%,固液比1.13 g·mL-1,pH 11.56,时间83 min,温度60℃条件下,提取率最大值为27.87%,醇溶蛋白纯度为92.47%,抗氧化试验表明,提取得到的醇溶蛋白对DDPH,ABTS和羟自由基有较强清除能力。Abstract: In order to extract high-quality zein, corn gluten meal were used as raw materials,ultrasonic-assisted extraction was applied to optimize the extraction process of zein by ethanol. A preliminary evaluation of antioxidant ability was finally conducted. The extraction process conditions were optimized through single factor experiment and response surface method experiment design. The results showed that when the ethanol concentration was 80%, the solid-liquid ratio was 1:13 g·mL-1, the pH was 11.56, the extraction time was 83 min, and the temperature was 60 ℃, the maximum extraction rate was 27.87%, the purity of extracted zein was 92.47%. The antioxidant test showed that the zein had strong scavenging ability towards DDPH, ABTS and hydroxyl free radicals.
