百合资源抗棉蚜性鉴定及遗传多样性分析

Identification of resistance to Aphis gossypii Glover and genetic diversity analysis of lily resources

  • 摘要: 鉴定百合资源抗棉蚜性和分析其遗传多样性,可为百合抗蚜及进一步抗病毒病育种提供重要亲本资源,提高百合抗蚜育种效率。连续3年,利用人工蚜虫接种和蚜量比值法鉴定60份百合资源(11份野生种、49份品种),荧光SSR标记分析遗传多样性。60份百合资源根据蚜量比值法可分为5级,12份资源为高感材料,6份资源为感虫材料,4份资源为中抗材料,7份资源为抗虫材料,31份资源为高抗材料。6对SSR引物共检测出35个主要等位变异,每个位点平均5.8个;多态性信息含量为0.5663~0.8940,平均0.7472;基因多样性指数变化为0.491~0.848,平均0.647;Shannon多样性指数为1.3759~2.5319,平均1.9231。不同抗蚜级别种质间遗传距离变化为0.1309~0.5403,平均0.3306,遗传一致度变化为0.5826~0.8773,平均0.7267。其中,感虫组和高感组资源遗传距离最小(0.1309),遗传一致度最大(0.8773),亲缘关系较近。UPGMA聚类分析结果表明,高抗百合组和中抗百合组资源聚为一类,高感百合组和感虫百合组资源聚为一类。个体间聚类,5组百合资源在多个组群中均出现。通过试验研究,建立百合种质资源抗棉蚜评价体系,明确部分资源抗蚜差异及遗传关系,筛选得到一批抗蚜种质资源,为百合抗蚜及进一步抗病毒病育种提供参考。

     

    Abstract: Identification of Aphis gossypii Glover resistance and analysis on genetic diversity of lily can provide important parental resources in lily aphid and virus resistance breeding and improve the efficiency of breeding. The 60 lily resources(11 wild species and 49 varieties) were artificially inoculated with aphids, the aphid number and its ratio were investigated from 2017 to 2019. Genetic diversity was analyzed based of fluorescently labeled SSR markers. The aphid resistance of lily could be divided into five grades in terms of aphid number ratio. Among 60 test resources, 12, 6, 4, 7 and 31 resources were highly susceptive, susceptive, moderate resistant, resistant and highly resistant. A total of 35 alleles were detected with an average of 5.8 alleles per locus with six SSR markers, the gene diversity index,polymorphism information content and Shannon’s information index ranged from 0.491 to 0.848, 0.5663 to 0.8940, 1.3759 to 2.5319, respectively, with an average of 0.647, 0.7472 and 1.9231, respectively.Genetic distance and genetic identity of accessions from different resistance groups were 0.1309 to0.5403(average value was 0.3306) and 0.5826 to 0.8773(average value was 0.7267), respectively, the smallest genetic distance(0.1309) was found between the sensibility group and the high sensibility group. UPGMA cluster analysis showed that the high resistance group and the moderate resistance group were clustered into one group and the high sensibility and sensibility groups were clustered into one group. The clustering result showed that the varieties(wild species) from five different resistance groups were divided into different groups. Through this study, the evaluation system of aphid resistance of lily resources was established, the genetic relationships of different aphid resistance resources were identified and the aphid resistance resources were screened out to provide useful reference for aphid resistance and further virus resistance breeding of lily.
