基于网络药理学和动物试验探究北五味子靶向PI3K/Akt信号通路发挥抗炎作用的机制

Study on anti-inflammatory mechanism of Schisandra chinensis targeting PI3K/Akt signaling pathway based on network pharmacology and animal experiment

  • 摘要: 为确定北五味子对动物抗炎的作用机制及效果,通过网络药理学分析北五味子发挥抗炎作用的有效成分、核心靶点和重要通路。利用葡聚糖硫酸钠诱导的溃疡性结肠炎小鼠模型分析北五味子抗炎活性及通路蛋白表达情况。结果表明,长栲利素A、脱氧三尖杉醋碱和当归酰基戈米辛O等8种成分为北五味子发挥抗炎作用有效活性成分,Pik3ca、Akt1、Map2k1等25个靶点为核心靶点,PI3K/Akt信号通路为重要通路。动物模型试验表明,北五味子可有效增加葡聚糖硫酸钠诱导的溃疡性结肠炎小鼠体重、结肠长度,降低结肠重量和肠重指数,调节促炎因子和抑炎因子间平衡,有效抑制结肠炎小鼠PI3K/Akt信号通路的活化发挥抗炎作用。

     

    Abstract: In order to determine the anti-inflammatory mechanism and effect of Schisandra chinensis on animals, network pharmacology was used to analyze the effective ingredients, core targets,and important pathways which Schisandra chinensis exerted anti-inflammatory effect. The dextran sulfate sodium-induced mice colitis model was utilized to assess the anti-inflammatory activities of Schisandra chinensis and the expression of pathway proteins. The results showed that eight substances were the active ingredients of Schisandra chinensis to exert anti-inflammatory effect, such as Longikaurin A,Deoxyharringtonine and Angeloylgomisin O. The anti-inflammatory effect could be attributed to the targeting of twenty-five core targets, including Pik3ca, Akt1 and Map2k1. The PI3K/Akt signaling pathway was a vital signaling pathway to excert anti-inflammatory effect. The animal model experiment had shown that Schisandra chinensis could effectively increase mouse body weight and colon length,reduce colon weight and colon-weight index, regulate the balance between pro-inflammatory and antiinflammatory factors in dextran sulfate sodium-induced mice colitis model, and effectively inhibited the activation of the PI3K/Akt signaling pathway.
