Abstract:
【Objective】 The study aimed to study genomic characteristics,pathogenicity and genetic evolution of Mycobacterium bovis CVCC68002,providing reference to the prevention and in-depth study of infectious diseases of Mycobacterium bovis.【Method】 The whole genome of CVCC68002 strain was sequenced by using Illumina and pacbio sequencing technology platforms,followed by establishing the library.The genomic function annotation analysis,pathogenicity and genetic evolution relationship analysis of the strain were performed by using bioinformatics and phylogenetic number.【Result】 The total genome length of the strain was 4 357 100 bp,with 65. 64% of GC,and there were 4 145 coding genes. The genomic characteristics of GO,KEGG,COG and NR databases showed that they had the functions in high amino acid and carbohydrate metabolism. The pathogenicity was analyzed through the databases of PHI,VFDB,ARDC and CARD,and the 456 virulence gene annotations were obtained,which were mainly related to bacterial adhesion and colonization,bacterial escape from the host immune defense system,secretory system proteins and transcription factors. The relevant drug-resistance genes,including vancomycin,penicillin,polyamine antibiotics,isoniazid,pyrazinamide,fluoroquinolone,etc.,were found by ARDB and CARD databases. In addition,the annotation amount of resistance protein of efflux pump was significant,and it was anticipated the bacteria could protect themselves by efflux of resistance protein.【Conclusion】 The complete genome information of Mycobacterium bovis CVCC68002 strain was obtained,and the phylogenetic information was improved. The virulence factors and effective target drug resistance genes were identified,which laid a theoretical foundation for the further study on molecular biological characteristics,pathogenicity and drug resistance gene screening of the strain.
