Abstract
In order to predict the role of Bcl-2 gene biological function in the cross regulation of apoptosis and autophagy, the structure and characteristics of the bovine Bcl-2 gene and the encoded protein were predicted by bioinformatics analysis using BioEdit,NCBI,ExPASy-ProtParam tool,TMHMM Server v.2.0,SPLIT 4.0,SignalP 4.1 Server,PSORTⅡ,PSIpred-MEMSAT 3 and SWISS–MODEL.The results showed that bovine Bcl-2 gene can encode 229 amino acid residues of soluble acid hydrophilic protein. The formula of protein is C1 132H1 717N311O316S9,the total number of atoms is 3 485,the theoretical isoelectric point is 6.65,the half-life is 30 h,the instability index is 58.20,the classification is unstable protein, the fat index is 76.77,the average value of hydrophilicity (GRAVY) is-0.133,and the prediction of Bcl-2 protein is hydrophilic protein;it contains transmembrane region and does not exist signal peptide;it is non secretory transmembrane protein;it is distributed in cytoplasm, nucleus, endoplasmic reticulum,Golgi, mitochondria, vesicles of secretory system, cytoskeleton, and vacuole;it is mainly composed ofα-helix(51.53%),extension chain(6.55%),β-angle(6.99%)and self-contained curl(34.93%),and its predicted results of the third-order structure are basically consistent with those of the second-order structure.Bovine Bcl-2 gene is highly conserved in mammals.Bcl-2 protein is mainly distributed in the cell membrane structure. It is an acidic hydrophilic transmembrane structural protein with hydrophobic structure at C-terminal,which is closely related to the regulation of apoptosis.
